In this paper, a novel highly-selective NH3 sensor based on a potassium trisoxalateferrate(III) complex was fabricated. Its gas-sensing properties were first investigated to NH3, H2S, NO2, SO2 and some organic solvent vapors. The sensors exhibited a high response to NH3 in a wide temperature range from room temperature to 150 degrees C. No responses were observed to H2S, NO2, SO2 and organic solvent vapors. The investigation proved that the response to NH3 was variable with the operating voltage. The optimum operating voltage was 5 V The response and recovery time were about 1.8 and 1.5 min, respectively. Since this sensor has an excellent selectivity, it appears promising for a range of gas-sensing applications. (c) 2007 Elsevier B.V. All rights reserved.

Cytoplasmic male sterility (CMS), which eliminates the possibility of self-pollination, is a commonly observed phenomenon in the plant kingdom and has been exploited for hybrid seed production. Rice (Oryza sativa L.) is one of the most important food crops in which CMS has been used successfully for commercial hybrid seed production. To better understand the molecular mechanism of CMS at the protein level during the development of anthers and microspores, proteomic analysis was performed. This technique was use to investigate the protein expression profile in tetrad-stage anthers of the rice CMS line, its fertile maintainer line, and the hybrid F1. Total anther proteins were extracted and separated by two-dimensional gel electrophoresis (2-DE). About 1,500 protein spots, including 49 differentially accumulated spots, were detected reproducibly and characterised by mass spectrometric analysis and database searches, and were identified as 48 unique proteins. Functional classification of the 48 differentially-expressed proteins indicated their involvement in several processes essential to pollen-development. These included metabolism, protein biosynthesis, transcription, signal transduction, cell death and disease responses, energy production, and cellular transporters. The potential roles of several differentially expressed proteins in CMS and fertile lines are discussed.

采用固相pH梯度/SDS-PAGE 双向电泳对红莲型细胞质雄性不育水稻的不育系(YTA)和保持系(YTB)二核期花粉总蛋白质进行了分离,通过银染显色,获得了分辨率和重复性较好的双向电泳图谱.用PDQuest 2DE软件可识别约1500个蛋白质点,其中差异表达的蛋白质点数为120.将其中15个差异点采用基质辅助激光解析电离飞行时间质谱(matrix assisted laser desorption/ionizaton time of flight mass spectrometry,MALDI-TOF-MS)进行了肽质指纹图分析,通过采用Mascot 软件对MSDB数据库查询,其中7个蛋白质点得到了鉴定.YTA相对于YTB有部分参与物质和能量代谢的蛋白质缺失或表达量降低,这些蛋白质分别是水稻线粒体H+-转运ATPase(H+-ATPase)α链、盐诱导型膜联蛋白、线粒体NAD+-依赖型苹果酶和磷酸核糖焦磷酸合成酶等.这些蛋白质的表达下调或缺失可能与线粒体提供能量不足而导致的花粉不能正常发育有关.线粒体电压依赖性阴离子通道(VDAC)这一重要蛋白质在YTA中的上调表达有可能与花粉败育过程中细胞的程序性死亡相关.

采用固相pH梯度-SDSPAGE双向电泳对红莲型细胞质雄性不育水稻的不育系(YTA)和保持系(YTB)单核期花粉总蛋白质进行了分离,通过银染显色,获得了分辨率和重复性较好的双向电泳图谱。Image Master 2D V5.0软件可识别约1800个蛋白质点,其中差异表达的蛋白质点数为85。将其中16个差异点采用基质辅助激光解析电离飞行时间质谱(matrix assisted laser desorption/ionizaton time of flight mass spectrometry,MALDI-TOF-MS)进行了肽质指纹图分析,通过采用Mascot软件对MSDB数据库查询,其中9个蛋白质点得到了鉴定。YTA相对于YTB有部分参与碳代谢和淀粉合成的酶缺失或表达量降低,这些蛋白质分别是ADP-葡萄糖磷酸转移酶(AGPase),UDP-葡萄糖醛酸脱羧酶,乙酰辅酶A合成酶和二氢硫辛酸脱氢酶等。其中AGPase是参与淀粉合成的蛋白,与花粉发育密切相关。乙酰辅酶A合成酶和二氢硫辛酸脱氢酶是细胞内合成乙酰辅酶A的重要酶,而乙酰辅酶A是进入TCA循环的重要底物,乙酰辅酶A的缺乏可以导致TCA循环不能顺利进行,从而不能提供小孢子发育所需要的大量能量。YTA相对于YTB部分参与碳水化合物代谢的重要酶缺失或表达量降低,有可能导致因线粒体提供的能量不足,淀粉合成受阻,因而花粉不能正常发育。
