Herein, DNA-templated copper nanoclusters (CuNCs) synthesized by terminal deoxynucleotidyl transferase (TdT) isothermal nucleic acid amplification (INAA) were proposed for Hg2+ assay. TdT recognized the 3′-OH end of primers and subsequently prolonged the single strand poly-T at 3′-OH with the assistance of deoxythymidine triphosphate (dTTP). The generated products coordinated with the copper ions to form DNA-templated CuNCs, resulting in remarkable fluorescence. The introduction of Hg2+ made the primers form a T-Hg2+-T mismatch duplex, which prevented the capture of the 3′-OH end by TdT. The poly-T single strand prolongation reaction was suspended; this led to fluorescence quenching. Hg2+ could be detected at a concentration as low as 76 pM with a significant linear range from 0.2 to 500 nM. This method, performed in one tube in one step, does not involve washing and separation steps, which provides a novel insight into the ultra-sensitive detection of toxic analytes in water quality applications. © 2019 The Royal Society of Chemistry.

MicroRNAs (miRNAs) may regulate diverse biological processes and play an important role in cancer. And MiRNAs have been proposed as a useful tool for lung cancer diagnosis and therapeutics in cancer. The purpose of the present study was to investigate the association among the expression level of mature miR-200b-5p in peripheral blood and the risk of lung cancer and clinic pathological characteristics. This case-control study included 24 patients with lung cancer and 12 healthy controls. MiR-200b expression was deleted using real-time PCR. and the miR-200b expression of normal controls was significantly higher than that in lung cancer patients (1732.13 pg/mL vs 881.67 pg/mL, P < 0.05), no difference with age, sex, tissue type and clinical stage of lung cancer patients (P > 0.05). Furthermore, miR-200b expression level fluctuated with tumor progression in lung cancer, and there was highly significant for clinical stage II compared with the clinical stage III (P < 0.05). In addition, the down-regulation of miR-200b showed a highly discriminative receiver operating characteristic (ROC) curve profile, clearly distinguishing cancer patients from cancer-free subjects with an area under the ROC curve (AUROC) of 0.87. The detection of miR-200b expression yielded 83.30% sensitivity and 100.00% specificity in the diagnosis of lung cancer. Therefore, these findings suggested that miR-200b may be used as a marker for the detection and diagnosis of lung cancer in peripheral blood. Background: Matrix metalloproteinases (MMPs) are capable of degrading and modifying most components of the extracellular matrix (ECM) and the basal membrane (BM), and play crucial roles in cancer invasion and metastasis. MMP gene expressions were regulated primarily at the transcriptional level, which was associated with tumor spread and patient prognosis. Polymorphisms in MMPs have been reported to be associated with non-small cell lung cancer (NSCLC). The objective of this study aim to evaluate the serum levels and polymorphisms of MMP-9 and MMP-13 in non-small cell lung cancer patients compared to normal subjects and their correlation to non-small cell lung cancer histopathology findings in Southern Chinese people. Methods: This case–control study included 245 patients with NSCLC and 258 healthy controls. Genomic DNA was extracted by using DNA extraction kit, genotyping was confirmed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and direct DNA sequencing, and serum levels of MMP-9 and MMP-13 were measured by using a specific ELISA, Human Matrix Metalloproteinase Enzyme Immunoassay Kits. Statistical analysis was carried out using the SPSS 23.0 software package. Results: The subjects carrying the TT genotype had a decreased risk of lung cancer in MMP9-1562C/T comparing with the CC genotype (p = 0.00, OR = 0.45, 95% CI = 0.29–0.68), and the MMP13-77 AA genotype was associated with a decreased risk of NSCLC by comparing with the GG genotype (p = 0.03, OR = 0.56, 95% CI = 0.33–0.94). Moreover, the C allele of MMP9-1562C/T could increase serum level of NSCLC in compared with the A allele (OR = 1.19, 95% CI = 0.75–1.89). Similarly, the AA genotype of MMP13 might be a marker of decreased serum level of lung cancer (OR = 0.76, 95% CI = 0.51–1.14). Conclusions: The results of these analyses underline the support of the notion that the CC genotype of MMP9-1562C/T and GG genotypes of MMP13-77G/A were associated with the increased risk NSCLC, and the serum levels of MMP9 and MMP13 were consistent with the results of the SNP analysis. MMP13 and MMP9 might be function as a key oncogene in NSCLC with a Southern Chinese population. Combined detection of SNP and enzyme activity between MMP9 and MMP13 are expected to be a potential diagnostic method of non-small cell lung cancer.

摘要:
Nanostarch has attracted much research interest recently due to its biodegradability and biocompatibility. A type of biocomposite film based on corn nanostarch (CNS) as the matrix and cellulose nanocrystals (CNC) as the reinforcement was prepared using a solution casting method. The influences of corn nanostarch concentration (CCNS), glycerin dosage (Dg), and cellulose nanocrystals dosage (DCNC) on the tensile strength of the biocomposite film were investigated by central composite design. The results were examined by an analysis of variance (ANOVA) and response surface methodology (RSM). The optimized process conditions as follows: CCNS of 11.25%, Dg of 12.00%, and DCNC of 5.00%. The CNS/CNC biocomposite film produced under these conditions showed a high tensile strength of 12.90 MPa. The CNS/CNC biocomposite film was characterized by Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), water contact angle, and scanning electron microscopy (SEM). The CNS/CNC biocomposite film has potential application prospects in the field of food and biomedical packaging. © 2019, North Carolina State University.

摘要:
The aim of present study was to evaluate the influence of cold storage on composition and properties of grass carp protein. The results show that muscle proteins of grass carp were degraded and underwent conformational changes when stored at 4 °C, and significant changes of the proteins content, as shown by the SDS-PAGE fingerprint, was observed after 6 days of storage. Protein’s surface hydrophobicity and total SH content increased during the first 4 days and then decreased gradually up to 10 days. The Ca 2+ -ATPase activity of protein decreased gradually during the storage period. The high MW proteins easily degraded during the cold storage, and the proteins with low MW were relatively stable but still gradually degraded. So the key time point for cold storage of grass carp is approximately 4-6 days. Copyright 2019 Zoological Society of Pakistan.

The effects of cold storage (0 °C, 0–8 days) and salts (NaCl, 0.5–5%;NaNO<inf>2</inf>, 50–150 mg/kg) on the formation of protein-bound N<sup>ɛ</sup>-(carboxymethyl)lysine (CML) and N<sup>ɛ</sup>-(carboxyethyl)lysine (CEL) in raw and subsequently commercially sterilized (121 °C, 10 min) pork were investigated. Based upon two-factor analysis of variance (α= 0.05), there was no significant interaction effect between salts and storage time on the levels of CML and CEL in raw or sterile pork;salts and storage time did not significantly affect the amounts of CML and CEL in raw pork. However, in sterile pork, 1.5–5% NaCl treatments led to an average of 7.5–52.6% increase in CML, while NaNO<inf>2</inf>resulted in 25.4–37.1% and 21.4–23.4% reduction in CML and CEL, respectively;raw pork stored for 8 days led to 82.5% increase of CML and 32.6% increase of CEL in the sterile products (n = 30), as compared to those without cold storage.<br/> ©2018 Elsevier Ltd

摘要:
选择复合蛋白酶水解鲢鱼制酶解产物,以添加了商业抗冻剂(商抗,4%蔗糖+4%山梨醇)的鱼糜制品作对照,综合评价添加了鲢鱼酶解产物的鱼糜制品冻融前后品质及其蛋白质体外消化率的变化。试验结果表明:冻融后,鱼糜制品中添加2%的酶解产物可以增加鱼糜凝胶持水性,降低蒸煮损失,并且不会导致鱼糜制品带上酶解产物本身的淡黄色;同时,2%酶解产物添加组能有效减缓鱼糜制品凝胶强度、硬度和弹性的降低及其蛋白质消化率的下降程度;2%酶解产物添加组在冻融前后感官评分均较高,酶解产物的添加避免了商业抗冻剂给产品带来的明显甜味。研究表明,鲢鱼酶解产物可作为潜在的抗冻剂应用于鱼糜及其制品的工业化生产。

摘要:
Cuprous oxide nanoparticles (Cu2O NPs) were dispersed into a graphene oxide (GO) solution to form a homogeneous Cu2O-GO dispersion. After this, the cuprous oxide nanoparticles were functionalized to electrochemically reduce the graphene oxide decorated glassy carbon electrode (Cu2O-ErGO/GCE). This was prepared by coating the Cu2O-GO dispersion onto the surface of the glassy carbon electrode (GCE), which was followed by a potentiostatic reduction process. An irreversible two-electron reaction of uric acid (UA) was observed at the voltammetric sensor. Moreover, the high concentrations of dopamine (DA) and ascorbic acid (AA) hardly affected the peak current of UA, which suggested that Cu2O-ErGO/GCE have excellent selectivity for UA. This is probably because the response peaks of the three compounds are well-separated from each other. The oxidation peak current was proportional to the concentration of UA in the ranges of 2.0 nM−0.6 µM and 0.6 µM−10 µM, respectively, with a low limit of detection (S/N = 3, 1.0 nM) after an accumulation time of 120 s. Cu2O-ErGO/GCE was utilized for the rapid detection of UA in human blood serum and urine samples with satisfactory results. © 2018 by the authors. Licensee MDPI, Basel, Switzerland.

摘要:
Catalytic degradation of organic pollutants by nanomaterials is an effective way for environmental remediation. The Fenton reaction involving H2O2 oxidation catalysed by Fe3+ is an advisable way for wastewater degradation. Herein, Fe3O4/SiO2 core-shell nanoparticles were prepared as catalyst by coprecipitation and sol-gel methods, and this catalyst is used for degradation of fuchsin in wastewater by H2O2. The Fenton reaction between H2O2 and Fe3O4 is proposed to explain the catalytic performance. The coating of SiO2 on Fe3O4 nanoparticles could dramatically stabilize the Fe3O4 in aqueous solution and prevent their oxidation. More importantly, the magnetic property of Fe3O4 nanoparticles endows them with good recyclability. Thus, due to the outstanding catalytic results, almost 100% removal degradation was achieved within 5 min over a wide pH value range at room temperature, which is better than that without catalysts. Temperature is a positive factor for improving the degradation rate, but room temperature is selected as the best temperature for economic and energy savings reasons, because more than 98% of fuchsins can still be degraded at room temperature. Moreover, these Fe3O4/SiO2 core-shell nanoparticles exhibit excellent magnetic recyclability and stable properties after repeated utilization. Therefore, these as-presented Fe3O4/SiO2 core-shell nanoparticles with low-cost and high performance are expected to be applied in practical industry wastewater degradation. © 2018 MDPI AG. 摘要:
通过对比分析经0~6次冻融循环处理后面团的酵母菌存活率、发酵力保存率、醒发时间以及焙烤性质研究了鲢鱼酶解产物和鱼胶原肽对冷冻面团品质的影响。结果表明:添加鲢鱼酶解产物和鱼胶原肽的面团经6次冻融循环后,酵母菌的存活率分别为78.95%和61.80%,发酵力保存率分别为59.06%和46.67%,面团醒发时间分别为330 min和318min,均明显优于空白组;冻融后的面团与新鲜面团相比,焙烤的面包比容和弹性减小,添加了鲢鱼酶解产物和胶原肽不仅可缓解冷冻面团这一缺陷,还可改善面包色泽。结果证明了鲢鱼酶解产物和鱼胶原肽具有良好的酵母菌抗冻保护活性。

以玉米纳米淀粉为基质,甘油为增塑剂、纤维素纳米晶(CNC)为增强剂采用流延成膜法制备玉米纳米淀粉/CNC复合膜,成膜基质和干燥温度对纳米淀粉成膜影响较大,聚氯乙烯基质板对纳米淀粉成膜较好,干燥温度25℃,成膜平整光滑;单因素探讨了玉米纳米淀粉、甘油和CNC含量对纳米淀粉/CNC复合膜强度性能的影响,在该基础上进行三因素三水平正交试验,正交优化研究表明,对玉米纳米淀粉/ CNC复合膜的抗张强度影响为:CNC>甘油>玉米纳米淀粉,正交试验优化结果为CNC 2%,甘油8%,玉米纳米淀粉10%,制备的玉米纳米淀粉/CNC复合膜抗张强度达20.18MPa;FTIR分析表明玉米纳米淀粉、甘油、CNC混合均匀,形成了均一稳定的纳米淀粉/CNC复合膜。该玉米纳米淀粉/CNC复合膜在食品药品可食性包装领域具有较好的应用前景。

摘要:
Contamination of water by meat production is an important and extensive environmental problem and even threat to human health. Biodegradation is a major mechanism which removes the pollutants from the environment. Therefore, the present study aimed to isolate and characterize a COD degrading bacteria which can effectively degrade slaughter wastewater. Six COD degrading bacteria were isolated from slaughtering waste water and sludge in Hunan a meat product Co., Ltd. And the COD degradation rate of each strain was determined by potassium permanganate method. Through observing morphologically and analyzing sequence to 16S rDNA, the highest COD degradation strain was Bacillus velezensis by preliminarily identified and classified, reaching 11.80%. The suitable conditions of the growth of Bacillus velezensis strain were 37 degrees C, pH 7.0, the peptone concentration 1.5%, and the yeast extract concentration 0.8%. Nanostarch has a small particle size and large surface area compared with traditional modified starch. In this study, nanostarch was prepared by dual screw extrusion and was characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), and X-ray diffraction (XRD). The nanostarch was studied as a coating adhesive in surface coating of lightweight coated paper. The crystallinity of nanostarch granules was decreased by dual screw extrusion, and its average particle size was about 100 nm. The results showed that for the paper coating formula of styrene butadiene latex and an ordinary oxidized coating starch ratio of 9:5, all ordinary oxidized coating starch and part of the styrene butadiene latex can be replaced by nanostarch. When 5 parts of ordinary oxidized coating starch and 4 parts of styrene butadiene latex were replaced by 6.5 parts nanostarch, the water retention property of nanostarch coating was increased by 64.0%, and its viscosity was decreased by 11.0%. The IGT printing surface strength of lightweight coated paper by the nanostarch coating was increased by 11.6%, and the glossiness of coated paper was increased by 7.2%. These results show that nanostarch surface coatings have good prospects for application in papermaking.

摘要:
优选酵母抗冻剂对改善冷冻面团品质具有重要意义。通过酶法制备了系列鲢鱼酶解产物,分析其基本组成和电泳图谱变化,并考察酶解时间及其浓度对面包酵母冻融过程中存活率的影响。结果表明:面包酵母耐冻性差,1个冻融循环后存活率仅为0.69%;不同酶解时间的鲢鱼酶解产物基本组成相差不大,但酶解15,30 min的酶解产物对酵母菌的抗冻保护效果最好;添加4%的30 min酶解产物使酵母菌的冷冻存活率提高到90.73%,较8%的甘油提高了42.88%。Tricine-SDS-PAGE分析发现,酶解产物的蛋白条带分布基本相似,但信号随着酶解时间的延长逐渐减弱,其中新增约13 kDa的蛋白可能与酶解产物的抗冻保护活性有重要关系。

To ensure food quality and safety, developing cost-effective, rapid and precision analytical techniques for quantitative detection of nitrite is highly desirable. Herein, a novel electrochemical sensor based on the sodium cellulose sulfate/poly (dimethyl diallyl ammonium chloride) (NaCS/PDMDAAC) composite film modified glass carbon electrode (NaCS/PDMDAAC/GCE) was proposed toward the detection of nitrite at sub-micromolar level, aiming to make full use of the inherent properties of individual component (biocompatible, low cost, good electrical conductivity for PDMDAAC; non-toxic, abundant raw materials, good film forming ability for NaCS) and synergistic enhancement effect. The NaCS/PDMDAAC/GCE was fabricated by a simple drop-casting method. Electrochemical behaviors of nitrite at NaCS/PDMDAAC/GCE were investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under optimum conditions, the NaCS/PDMDAAC/GCE exhibits a wide linear response region of 4.0 × 10-8 mol.L-1~1.5 × 10-4 mol.L-1 and a low detection 1imit of 43 nmol.L-1. The NaCS/PDMDAAC shows a synergetic enhancement effect toward the oxidation of nitrite, and the sensing performance is much better than the previous reports. Moreover, the NaCS/PDMDAAC also shows good stability and reproducibility. The NaCS/PDMDAAC/GCE was successfully applied to the determination of nitrite in ham sausage with satisfactory results. © 2018 by the authors.

Human epidermal growth factor receptor 2 (HER2) is a key tumor marker for several common and deadly cancers. It is of great importance to develop efficient detection methods for its over-expression. In this work, an electrochemical impedance spectroscopy (EIS) method adjustable by anionic porphyrin for HER2 gene detection has been proposed, based on the impedance difference between multi-walled carbon nanotubes (MWCNTs) and DNA. The interesting finding herein is that with the addition of anionic porphyrin, i.e., meso-Tetra(4-sulfophenyl)-porphyrin (TSPP), the impedance value obtained at a glass carbon electrode (GCE) modified with MWCNTs and a single stranded DNA (ssDNA), the probe DNA that might be assembled tightly onto MWCNTs through φ φacking interaction, gets a slight decrease; however, the impedance value from a GCE modified with MWCNTs and a double stranded DNA (dsDNA), the hybrid of the probe DNA with a target DNA, which might be assembled loosely onto MWCNTs for the screening effect of phosphate backbones in dsDNA, gets an obvious decrease. The reason may be that on the one hand, being rich in negative sulfonate groups, TSPP will try to push DNA far away from CNTs surface due to its strong electrostatic repulsion towards DNA; on the other hand, rich in planar phenyl or pyrrole rings, TSPP will compete with DNA for the surface of CNTs since it can also be assembled onto CNTs through conjugative interactions. In this way, the ""loosely assembled"" dsDNA will be repelled by this anionic porphyrin and released off CNTs surface much more than the ""tightly assembled"" ssDNA, leading to a bigger difference in the impedance value between dsDNA and ssDNA. Thus, through the amplification effect of TSPP on the impedance difference, the perfectly matched target DNA could be easily determined by EIS without any label. Under the optimized experimental conditions, this electrochemical sensor shows an excellent linear response to target DNA in a concentration range of 2.0 &times10-11-2.0 &times; 10-6 M with a limit of detection (LOD) of 6.34 ×10-11 M (S/N = 3). This abnormally sensitive electrochemical sensing performance resulting from anionic porphyrin for DNA sequences specific to HER2 gene will offer considerable promise for tumor diagnosis and treatment. © 2018 by the authors.

A facile cuprous oxide nanoparticles functionalized electro-reduced graphene oxide modified glassy carbon electrode (denoted as Cu 2 O NPs-ERGO/GCE) was fabricated via a simple physical adsorption and electrochemical reduction approach. Cyclic voltammetry and second-order derivative linear scan voltammetry were used to investigate the electrocatalysis oxidation of vanillin on the Cu 2 O NPs-ERGO/GCE. The compound yielded a well-defined voltammetric response in 0.1 M H 2 SO 4 at 0.916 V (vs. saturated calomel electrode (SCE)). A linear calibration graph was obtained in the concentration range of 0.1 μM to 10 μM and 10 μM to 100 μM, while the detection limit (S/N = 3) is 10 nM. In addition, the Cu 2 O NPs-ERGO/GCE presented well anti-interference ability, stability, and reproducibility. It was used to detect vanillin sensitively and rapidly in different commercial food products, and the results were in agreement with the values obtained by high performance liquid chromatography. © 2018 by the authors. 摘要:
分析了鲢鱼加工副产物酶解产物对冻融鱼糜肌原纤维蛋白性质的影响。采用复合蛋白酶制备酶解产物,分别添加2%,6%的酶解产物(FPH-2%、FPH-6%)于鱼糜中,设置空白对照,冻融循环6次后,提取鱼糜中的肌原纤维蛋白并测定其性质。结果表明:两种添加量的酶解产物对冻融鱼糜中肌原纤维蛋白均具有良好的保护作用,其中,添加6%酶解产物的鱼糜冻融循环6次后,肌原纤维蛋白浓度和巯基含量降低最少且羰基含量较低,DSC和SDS-PAGE分析表明:6%酶解产物添加组中肌原纤维蛋白变性程度较低。本研究有助于明确该酶解产物对于鱼糜结构的作用,为鲢鱼加工业提供理论依据;同时能有效利用蛋白资源,提高水产加工产业的附加值。
